Does a hole inside of a hole make the second hole "holier?"

Yeah I know I have a hole inside of my head.
I'm trying to get at the exact meaning of a hole. What do we mean by it? The question may sound silly, but silly questions can lead to progress (and can lead to a lot of fun).
So have a go at it and see what you can come up with.

In Italy we have candy, mints, Polo. The ad says: Polo, the hole with the mint around.

We have, on the basis does a hole exist inside a hole? This time looking at what would make a hole a hole so I'm using a ridiculous example to examine the question.

no a hole inside hole just means stop digging. In maths the hole is just a hole to which the number is always the same regardless of the whole.
But if you must:

Eels. they had a point, even with a polo on their face, elements of either not quite combining to make them as good as either, wise words in logic. Calculus is your boring but quite apt friend on holes. Although it is interesting to note in set and number theory, every number and hence everything relies on nothing or zero.

No, it makes it a bigger or deeper hole.

Not necessarily. It could mean one hole is smaller.
